If you're asking if it is in relation to Yakuza the answer is no. Ishin is based off an actual Japanese folk tale. The game's just using Yakuza characters for the likeliness of the actual people being portrayed

Not a folk tale, it's real history. Well, a very fictionalised version of real history.

As to it being canon it wasn't but, in a very strange way, it kinda is now; in an off comment in Like A Dragon: Infinite Wealth Kiryu mentions having a dream of being Sakamoto Ryōma and even speculates that he might be experiencing real memories. So yeah, within the greater Yakuza canon this entire game is Kiryu's dream.
